I have a very simple question. Will Charged Gauntlets ignore the targets Defense chance when it procs, as it is an automatic critical hit, or can it still Miss/Dodge?

No. The rail shot can still miss. If it hits, it has a 100% chance to crit

Also keep in mind, if you're targeting an enemy, specifically an enemy player, that has some form of shield, Rail Shot fired when Charged Gauntlets is proc'd will not ignore that, so it will either be absorbed or have some form of reduction.
