It happens, there are several NPCs that have that as their back-story or as something that you can make happen to them based on your choices (a lot come up in the Knight storyline in particular). As a gameplay mechanic for your player characters? No. There are no faction changes/defection mechanics at all in the game. Even if you play extremely Light Side as an Empire Character or extremely Dark Side as a Republic Character, you stay affiliated with the faction you start on as far as gameplay goes.
